Abstract

This study aims to design an innovative flexible nanocomposite film via the integration of MnO2 NPs into the biopolymeric carboxymethyl cellulose (CMC) hydrogel for improving its mechanical, swelling, and antibiotic release profile. To end this, the casting method was utilized to incorporate MnO2 NPs in the biopolymeric CMC matrix (CMC/MnO2) plasticized by glycerol and cross-linked by citric acid. Also, colistin (CL) was loaded in the nanocomposite formulation owing to its antibacterial activity. The constructed films were characterized and confirmed by different methods. The release study of CL exhibited a low initial burst release with a controlled profile over 72 h at pH 7.4 simulating the wound media. Remarkably, the results of antibacterial and hemolysis assessments showed a significant activity toward E. coli bacteria with 1.9 cm inhibition zone, cytocompatibility against HFF-1 cell lines (over 85% cell viability), and a good hemolysis rate (about 5%) after incubation with human red blood cells. The results recommend CL-loaded CMC/MnO2 (CL/CMC/MnO2) nanocomposite film as a potent antimicrobial wound bandage.
