Abstract

Aim ofthisstudywastoinvestigatetheinvolvementofthemitochondrialpermeabilitytransitionpore (MPTP) andoxidativestressinthecardioprotectiveeffectofvasopressin(AVP)onischemia/reperfusion (I/R) injury.Anesthetizedmalewistarratsweresubjectedtoregional30minischemiaand120min reperfusion andrandomlydividedintoninegroups:(1)Control;salinewasadministeredintravenously before ischemia,(2)vasopressinwasadministrated10minpriortoischemia,(3,4)AtractylosideasMPTP opener,wasinjected5minpriortoreperfusionwithoutandwithvasopressin,(5,6)CyclosporineAasa MPTP closer,wasinjected5minpriortoreperfusionwithoutandwithvasopressin,(7)mitochondria wereisolatedfromcontrolgroupandCaCl2 wasaddedasMPTPopenerandswellinginducer,(8)isolated mitochondria fromControlheartswasincubatedwithCyclosporineAbeforeaddingtheCaCl2 (9) CaCl2 wasaddedtoisolatedmitochondriafromvasopressingroup.Infusionofvasopressindecreasedinfarct size (18.671.7%vs.controlgroup37.672.4%), biochemicalparameters[LDH(LactateDehydrogenase), CK-MB(CreatineKinase-MB)andMDA(Malondialdehyde)plasmalevels,PAB(Prooxidant–antioxidant balance)] comparedtocontrolgroup.Atactylosidesuppressedthecardioprotectiveeffectofvasopressin (32.571.9%vs.18.671.7%)butadministrationoftheCyclosporineAwithoutandwithvasopressin significantly reducedinfarctsizeto17.774% (Po0.001)and22.773% (Po0.01)respectively,vs. 37.672.4% incontrolgroup.Also,vasopressin,similartoCyclosporineA,ledtodecreaseinCaCl2- induced swelling.ItseemsthatvasopressinthroughantioxidanteffectandMPTPinhibitionhascreateda cardioprotectionagainstischemia/reperfusioninjuries.
